Establishment of the model of porcine acute liver failure

Abstract
AIM To provide a reliable animal model for the study of treatment of acute liver failure with bioartificial liver.
METHODS Blood of eight native hybrid pigs into the livers was obstructed completely by two-phase operations. Firstly, the pigs underwent potal-right renal venous shunt. And two days later, hepatic artery, gastroduodenal artery and peri-hepatic ligaments were cut to obstruct blood flow into the liver. Acute loss of compensation of liver functions was produced and the pigs died of acute liver failure. At definite intervals (every 2h after phase Ⅱ surgery), blood was collected to assay ALT, AST, TBil Alb, Ammonia, BUN, Cr, PT and Fib and liver tissues were sampled for pathological examinations under light microscopy and electronic microscopy.
