     模拟失重对大鼠骨肉瘤细胞内Ⅰ型胶原表达的影响

    【摘要】 目的 通过观察回转条件下培养的大鼠骨肉瘤细胞内Ⅰ型胶原的基因和蛋白表达的变化,探讨模拟失重对成骨细胞增殖功能的影响。 方法 对培养的大鼠骨肉瘤细胞采用回转器模拟失重24、48、72 h,同时设1 G对照组。在各时间点提取细胞总RNA,进行反转录PCR,检测Ⅰ型胶原α1链(COL-Iα1)mRNA的表达情况,同时采用ELISA法测定细胞内Ⅰ型胶原蛋白的表达量。 结果 模拟失重72 h细胞内COL-Iα1 mRNA和Ⅰ型胶原蛋白的表达量均明显低于对照组。 结论 回转器模拟失重72 h使大鼠骨肉瘤细胞的增殖功能降低。

    【Abstract】 Objective To investigate the effects of simulated weightlessness on the proliferation of osteoblastic cells through the change of expression of type I collagen in rat osteosarcoma cells cultured in the rotating clinostat. Method Three experimental groups of the cultured rat osteosarcoma cells underwent simulated weightlessness in rotating clinostat for 24, 48 and 72 h respectively. The control groups in 1 G condition were set up at the same time. The total RNA in cells was isolated and reverse transcription PCR analysis was made to examine the gene expression of collagen Iα1. The type I collagen protein in cells was quantified by ELISA. Results The quantity of collagen Iα1 mRNA and type I collagen protein after 72 h exposure to simulated weightlessness was significantly less than that of control group. Conclusion The proliferation of rat osteosarcoma cells was retarded by 72 h rotating clinostat simulated weightlessness.
