对MGC-803胃癌细胞株癌基因表达及调控的实验研究

    【Abstract】 Objective To study the regulation of rTNF,rIL-6,rIL-2, 60 Co-rays on the expression of the genes of P53,APC and c-Ha-ras by using human gastric carcinoma cell line MGC803as the model.Methods The expression levels of P53,APC and c-Ha-ras mRNA of human gastric carcinoma cell line MGC803were detected by in situ hybridization technique,and the expression level of the c-fos protein was detected through immunohistocheˉmical SP(Stretavidin/Peroxidase)method.Results It was found that human gastric carcinoma cell line MGC803can’t express P53and APC,but can express C-Ha-ras and c-fos highly.The results were shown as follows:The expression of P53was induced P<0.01;The expressions of c-Ha-ras and c-fos were inhibited P<0.01by rTˉNF;The expressions of c-fos was inhibited rIL-6P<0.01;The expressions of C-fos was inhibited rIL-2P<0.01.There were no genetic expressions in the early stage of irradiation(6h),however,there were expressions of c-Ha-ras and c-fos in various degrees during the late stage.Conclusion There is the regulation of rTNF ......
