【摘要】 目的 构建表达带pre S的乙肝表面抗原的质粒，并在真核细胞CHO中表达该抗原。方法 通过PCR方法分别扩增乙肝病毒表面抗原S和前S1及S2基因，利用引物的柔性短肽连接头，经第二次PCR扩增，获得可完整表达乙肝病毒表面抗原的融合基因。分别以pcDNA3和p SV2载体构建真核细胞表达质粒，共转染CHO细胞，表达产物用特定抗原酶联免疫检测和反向被动血凝实验检测。结果 表明表达物具有完整乙肝病毒表面抗原特性。结论 成功地构建了含前S基因的重组乙肝病毒表面抗原表达质粒，并在CHO细胞中表达。
