高压氧对局灶脑缺血细胞凋亡的影响及其作用机制

     【摘要】 目的 探讨高压氧(HBO)对持续性局灶脑缺血细胞凋亡的影响及其作用机制。方法 选择4~5个月龄SD大鼠65只,随机分组制作持续性局灶脑缺血动物模型组(缺血组)、缺血高压氧治疗组(HBO组)和假手术组(对照组)。观察脑梗死灶体积、凋亡细胞数、凋亡调控基因蛋白表达的变化。结果 HBO组与缺血组相比:6h~10d各相应时间点梗死灶体积都明显缩小,差异有非常显著性(均P<0.01);HBO组48hTUNEL阳性凋亡细胞数较缺血组显著减少(P<0.01);HBO组与缺血组相比,在6h~10d各时间点Bcl-2免疫组化阳性细胞增加,差异有显著性(P<0.05);Bax免疫组化阳性细胞在6h~10d各时间点均减少,差异有显著性(P<0.05)。结论 脑缺血早期暴露于高压氧可以减少局灶脑缺血梗死灶体积;高压氧能抑制脑缺血细胞凋亡,其作用与上调Bcl-2蛋白、下调Bax蛋白表达有关。

    【Abstract】 Objective To study the effect of hyperbaric oxygen(HBO)on apoptosis in focal cerebral isˉchemia and mechanism.Methods Sixty five4-5-month-old Spraque-Dawley(SD)rats were divided into three groups(focal cerebral ischemia group,FCI group,HBO group and control group)at random.To detect the focal brain tissue infarct sizes(BTIS),TUNEL numbers and expression of bcl-2and Bax protein.Results The BTIS are signifˉicantly reduced from6hoursto10days and the TUNEL numbers are significantly decreased after HBO48hours in HBO group(P<0.01).The bcl-2protein levels are increased significantly and Bax protein levels are significantly decreased in HBO group(all P<0.01).Conclusion HBO can reduce focal brain tissue infarct sizes and inhibit apoptosis in early treatment focal cerebral ischemia.Its mechanism may be related to the up-regulation of bcl-2proˉtein levels and down-regulation Bax protein levels. ......
