深低温冻存软骨细胞体外立体培养的生物学特性研究

     【摘要】 目的 研究深低温冻存对软骨细胞生物学特性的影响。方法 取经深低温冻存1、2、3个月的软骨细胞、复苏后体外培养,取第3代对数生长期培养细胞,制成细胞悬液,调整其为5×10 7 个/ml左右,接种于PGA三维支架材料上,复合物体外培养2周,行电镜及组织学观察。结果 经深低温冻存的软骨细胞与新鲜的软骨细胞一样,在复合物体外培养1周,细胞开始出现分裂增殖,细胞周围集聚异染性细胞外基质。结论 经深低温冻存的软骨细胞保持了分裂增殖及合成基质的能力,可用于组织工程构建组织工程化软骨。

    The study of biological character of tridimensional cultured chondrocytes after cryopreservation in vitro

    【Abstract】 Objective To study the effect of biological characteristics of cryogenically frozen chondrocytes.Methods Cryogenically frozen chondrocytes were thawed and cultured in a monolayer in serum-based chondrocyte meˉdia.They were seeded onto3-dimensional biopolymer scaffolds in a spinner flask for2weeks.Chondrocyte growth and extracellular matrix production in the constructs were confirmed by cell count,cell viability and histologic analysis and by electron microscopy.Results Histologic sections confirmed the presence of chondrocyte growth and extracellular matrix production in the constructs.There was no difference in cryopreserved and noncryopreserved chondrocytes.Conclusion This study proves that cryogenically preserved chondrocytes retain their ability to grow in tissue culture ......
