AS扩血管机制的研究

     【摘要】 目的 应用大鼠血管平滑肌等张收缩的原理和放射免疫测定环磷酸核苷酸水平等方法,对阿魏硝胺(AS)的扩血管作用的机制进行探讨。方法 用硝酸甘油作对照观察AS对去甲肾上腺素和高钾预收缩血管的松弛作用;检测AS对大鼠胸主动脉条生成cAMP及cGMP的影响。结果 AS松弛去甲肾上腺素(1μmol/L)和氯化钾(30mmol/L)预收缩的大鼠胸主动脉条,其EC 50 分别为(35.9±3.1)和(36.7±3.1)μmol/L。硝酸甘油的EC 50 分别为(18.6±3.8)和(18.1±2.2)μmol/L。AS显著增加大鼠胸主动脉血管平滑肌内cGMP水平(P<0.01)。结论 AS通过提供外源性NO · 、提高平滑肌细胞内cGMP水平,从而产生血管扩张作用。
