摘要 目的:探讨米非司酮对人卵巢上皮性癌细胞的 杀伤活性及对细胞增殖和细胞周期分布的影响。方 法:采用MTT测定,观察不同浓度米非司酮(20、10、5、 2.5μmol/L)对HO-8910细胞的杀伤活性;通过ABC免疫组化 染色及流式细胞仪分析,检测用药后HO-8910细胞Ki-67抗 原表达、细胞周期分布及细胞增殖指数的变化。结 果:4个浓度的米非司酮对HO-8910细胞均有杀伤作用, 并呈剂量依赖性,以20μmol/L浓度杀伤作用最明显,杀 伤率达52.01%;5μmol/L米非司酮可使HO-8910细胞Ki-67抗 原表达显著减少,细胞增殖受到明显抑制,使细胞 阻滞于G0/G1期,不能进入S及G2/M期,细胞增殖指数明显 下降。结论:米非司酮体外对人卵巢上皮性癌细胞 具有杀伤活性,并对细胞增殖及细胞周期有明显抑 制作用,主要通过抑制DNA合成,将细胞阻滞于G0/G1期 。

Effects of Mifepristone on Cell Proliferation and Cell Cycle Kinetics of Human Ovarian Epithelial Carcinoma Cell Line HO- 8910 in Vitro

Abstract Objective: To study the effects of mifepristone on cell proliferation and cell cycle kinetics of human ovarian carcinoma cell lines HO- 8910 in vitro. Methods: The antiproliferation effects of four different concentrations of mifepristone (20,10,5,2.5μ mol/L) on HO- 8910 cell line were determined with MTT assay. Immunohistochemical (ABC) and flow cytometry techniques were used to determine the changes of Ki- 67 antigen expression and cell cycle distribution and cell proliferation index after treatment with mifepristone. Results: Four different concentrations dose- dependently inhibited the proliferation of HO- 8910 cells. The inhibition by mifepristone plateaued at 20μ mol/L (52% inhibition). Mifepristone at 5μ mol/L decreased the expression of Ki- 67 antigen of HO- 8910 cells. It (5μ mol/L) caused a decline in the proportion of cells in S and G2/M phase, blocked the cell cycle progression in G0/G1 phase and reduced the proliferation index (PI) of HO- 8910 cells. Conclusion: In vitro, Mifepristone significantly inhibites the growth of human ovarian epithelial carcinoma cells which are progestin receptor positive and leads to arrest of the cell cycle progression in G0/G1 phase.
