【摘要】目的 建立微量反应板测定血小板聚集的方法(MP法).方法 血小板最佳浓度为200±50×10⁹ /L;ADP最佳浓度为4unmol/L(终浓度);酶标仪波长选择655mm;标本振荡用旋涡混合器.结果 批内精密度6.58%,与血小板聚集仪法同时对15例标本的血小板最大聚集率测定结果进行了比较,两法基本一致,无显著性差异,P>0.05.结论 MP法适合于临床及科研对批量血小板聚集率的测定。

Measuring Platelet Aggregation with Microplate Reader

【Abstract】Objective To develop the method of measuring platelet aggregation with microplate reader(MP).Methods The best concentration of platelet suspension is 200±50×10⁹ /L;The final concentration of ADP is 4unmol/L; wavelength 655nm;The sample volume 150ul,the sample is agitated by a vortex agitator between each reading cycle.Results The coefficiency of variation within (CV) is 6.58%.Compared with the results of 15 samples measured by microplate reader and aggregometer,there is no obvious difference.Conclusion The microplate reader is suitable for clinical determination and scientific research for a batch of platelet aggregation evaluations.
