【摘要】 目的 建立稳定、成熟的内皮细胞原位获取法。方法 应用原位获取法从恒河猴及成人的73条表浅或腹腔血管中获取内皮细胞。A、B两组分别用0.1%Ⅱ型胶原酶消化25分钟、15分钟。结果 A组内皮细胞获取率明显高于B组。两组中大隐静脉、颈外静脉、上肢浅静脉、胃网膜右静脉、髂总动脉的内皮细胞获取率明显高于既往的机械获取法、灌注酶解消化法、外翻酶解消化法(P<0.01)。结论 原位获取法的内皮细胞获取率明显高于既往的内皮细胞获取方法,其理想的消化时间25分钟。

    【Abstract】 Objective
To establish a new harvest method for in situvessel cannulation for endothelial cell harvest. Method 73 vessels ofmacacas and adults were incubated with 0.1% collagenase for 25 min (group A) or 15 min(group B). In situ vessel cannulation was used to harvest the endothelial cells (ECs) fromthe vessels. Result The harvesting rate of ECs in group A was much higherthan in group B. The harvesting rate of ECs in great saphenous vein, external jugularvein, peripheral super limb veins, right gastroepiploic vein and common iliac artery inboth group A and group B was increased obviously with this method as compared withprevious method. Conclusion The harvesting rate of ECs was increased byusing in situ harvest method and the ideal incubation time was 25 min.
