【摘要】 目的 探讨端粒酶在胃癌组织中的活性及其临床意义。方法 应用银染TRAP方法检测94例胃癌组织、12例胃腺瘤、9例胃溃疡和58例癌旁正常胃粘膜中端粒酶活性。结果 94例原发胃癌组织中检出端粒酶阳性81例，阳性率为86.2%。正常粘膜无1例检测到端粒酶表达，12例胃腺瘤有1例表达端粒酶阳性，在9例胃溃疡组织中有1例检测到端粒酶表达，12例胃腺瘤有1例表达端粒酶阳性，在9例胃溃疡组织中有1例检测到阳性端粒酶表达。胃癌与正常粘膜组端粒酶表达率差异有显著性(P<0.001)。端粒酶活性与肿瘤大小、分化程度、浸润深度、淋巴结转移和TNM分期无明显相关(P>0.05)。结论 测定胃癌端粒酶活性对胃癌的诊断有一定意义。

Telomerase activity in human gastric carcinomas

【Abstract】 Objective To studythe telomerase expression in gastric carcinomas and the clinical implications. Methods Telomerase activity was detected by a non-isotopic PCR-based TRAP assay (telomeric repeatamplification protocol) in tissue samples from 94 gastric carcinomas, 58 normal tissues,12 gastric adenomas and 9 gastric ulcer lesions.Results None of thenormal tissues exhibited telomerase activity. In contrast, 81 out of the 94 patients withcancers were positive for telomerase expression (P<0.001), 1 of 12 with gastric adenomasand 1 of 9 gastric ulcer also were positive for telomerase expression. The incidence oftelomerase activity in gastric carcinoma tissues was not correlated to tumor size,differentiation, tumor invasion in depth, lymph node metastasis or TNM stage (P>0.05).Conclusion Telomerase might be a good diagnostic marker for the diagnosis of gastric carcinoma.
