【摘要】目的探讨淋巴细胞因子激活的杀伤细胞/活化自然杀伤细胞(LAK细胞/活化NK细胞)与造血的关系及其对同基因骨髓移植小鼠造血重建的影响。方法①将同基因LAK细胞/活化NK细胞与骨髓细胞一起置于体外半固体琼脂培养体系中,观察其对CFUGM形成的影响。②在小鼠同基因骨髓移植中,联合注射同基因LAK细胞/活化NK细胞,观察其对移植后小鼠造血重建的影响。结果①当骨髓细胞置于有外源性集落刺激因子(CSF)的适宜培养体系中,同基因LAK细胞/活化NK细胞抑制CFU-GM形成;而当骨髓细胞置于无CSF不良培养体系中,同基因LAK细胞/活化NK细胞促进CFU-GM形成。②在小鼠同基因骨髓移植中,联合注射同基因LAK细胞/活化NK细胞,能显著促进CFU-S和CFU-GM增殖和显著升高白细胞和血小板数量。结论①同基因LAK细胞/活化NK细胞对CFUGM形成有双向调节作用。②在小鼠同基因骨髓移植中,联合注射同基因LAK细胞/活化NK细胞,能促进移植后小鼠造血重建。

Experimental study of LAK and activated NKcells

    affecting hematopoiesis in mice

Abstract Objective Tostudy the relationship between LAK/activated NK cells and hematopoiesis as well as theeffects of LAK/activated NK cells on hematopoietic reconstitution after syngeneic bonemarrow transplantation(BMT) in mice. Methods ① LAK/activated NK cellswere cocultured with bone marrow cells(BMC) in semi-solid agar medium to determine theireffects on CFU-GM formation. ② Syngeneic LAK/activated NK cells were transferred with BALB/c BMCs into lethally irradiated syngeneic recipients to determine their effects onhematopoietic reconstitution. Results When BMC were cultured underoptimal growth conditions in the presence of exogenous hematopoietic growth factors, theeffects of LAK/activated NK cells were mainly inhibitory. However, when BMCs were culturedunder growth-limiting conditions without exogenous hematopoietic growth factors, the LAK/activated NK cells could support CFU-GM formation. Syngeneic LAK/activated NK cellssignificantly increased CFU-S and CFU-GM numbers after syngeneic BMT,and alsosignificantly improved platelet and white blood cell count. Conclusions LAK/activated NKcells could be bidirectionally regulate CFU-GM formation in vitro and promotehematopoietic reconstitution after syngeneic BMT in mice.
