摘要 目的:探讨非洛地平对家兔血管成形术后内皮功能及内膜增殖的影响。方法:日本大耳白兔36只,高脂喂养4周后用球囊导管剥脱腹主动脉内皮,再高脂喂养4周后,对动脉粥样硬化部位行血管成形术,30只成功的动物随机分为对照组、高脂组和治疗组(非洛地平),继续喂养4周后采集局部血管行病理形态学分析,并观察实验前后各组血清脂质、一氧化氮及血浆内皮素-1的改变。结果:治疗组血管内膜面积与中膜面积之比及内膜面积均降低。各组高脂饮食后血清总胆固醇、甘油三酯和血浆内皮素-1均明显升高,而血清一氧化氮明显下降;12周治疗组血浆内皮素-1明显降低,而血清一氧化氮水平明显升高。结论:非洛地平具有抑制家兔血管成形术后内膜增殖和保护内皮功能的作用。

The Effects of Felodipineon Endothelial Function and Neointimal

    Proliferation After Angioplasty in Rabbits

Abstract Objective:To investigate theeffects of felodipine on endothelial function and neointimal proliferation afterangioplasty of the rabbits.Methods:The celiac arterial endothelium of 36 male Japanesewhite rabbits were denuded after being fed with high-cholesterol diet for 4 weeks,and theangioplasty to atherosclerotic stenosis of celial arteries was performed after 4 addtionalweeks of high-cholesterol diet.30 successful animals were randomized to the control group,high-cholesterol group and felodipine group.In continuous 4 weeks later,the localvessels were collected for morphological observation,and blood lipid.Nitric oxide (NO)andendothelin-1(ET-1)were also meassured during this experiment.Results:The morphologicalohservation showed that the neointimal area(NEA)and neointimal to medial area ratio(NEA/MA)in felodipine group were less than the control group,and those in controlgroup were less than in high-cholesterol group.The serum total cholesterol(TC) triglyceride (TG)and ET-1 levelswere increased and the serum NO level had a tendency to decrease after high-cholesteroldiet.However in comparision to the control group,there were apparently decreased TC TG and ET-1 levels and increasedNO level in felodipine group.Conclusion:The felodipine had the effects of inhibiting theneointimal proliferation and of protecting the endothelial function after angioplasty inrabbits.
