照射后小鼠小肠上皮细胞TGF-β及其受体TGF-β R1和TGF-β R2的表达变化及其意义

     【摘要】 目的 探讨TGF-β及其受体TGF-β R1和TGF-β R2在小鼠小肠上皮细胞受照射后再增殖中的作用及其意义。方法 C3HF/Kam小鼠全身一次性5Gy照射后,不同时间内杀死小鼠取出空肠,HE染色后计数隐窝细胞数和分裂相,同时以免疫组化结合计算机图像分析技术定量测定照射后不同时间内TGF-β、TGF-β R1和TGF-β R2在隐窝区和绒毛区的表达。结果 照射后3~4天小鼠小肠隐窝细胞和分裂相均达到高峰。而照射后一天TGF-β表达在绒毛区和隐窝区即下降至最低点;同时TGF-β R1和TGF-β R2也不同程度的下降。结论 TGF-β、TGF-β R1和TGF-β R2在小鼠小肠上皮细胞受照射后的再增殖过程中可能起着负反馈的调节作用。

    The role of expression of TGF-β,TGF-β and TGF-β R2 in the repopulation of mouse jejunum after irradiation

    【Abstract】 Objective To study the role of expressions of TGF-β,TGF-β R1 and TGF-β R2 in repopulation of jejunum following irradiation.Methods Using the jejunum of C3HF/Kam mouse as material,the mitotic index and the number of crypt cells were measured at different times after single whole body irradiation of 5Gy.At the same time,the expressions of TGF-β,TGF-β R1 and TGF-β R2 in crypt area and villi area were also tested quantitatively by automated immunohistochemical staining method.Results The mitotic index and the number of crypt cells reached peaks three-four days after irradiation.Meanwhile,the expression of TGF-β in crypt area and in villi area showed the lowest value one day after irradiation and the expreesions of TGF-β R1,2 decreased in some degrees both in crypt and in villi.Conclusion The results indicate that the decreased expression of TGF-β, TGF-β R2 play the negative feed back role in repopulation of mouse jejunum after irradiation. ......
