氧化苦参碱在大鼠离体小肠各肠段吸收的研究

    摘要:目的 选择大鼠各个肠段进行肠吸收试验,考察氧化苦参碱在大鼠小肠十二指肠、空肠、回肠及结肠部位的吸收情况,确定是否存在药物特异性吸收部位,以此来证明氧化苦参碱制备成缓释制剂的可能性。方法 采用大鼠离体小肠外翻肠囊法试验,用HPLC法检查肠液中药物浓度。结果 苦参素在2 h内空肠和回肠中的累积吸收量最大,在十二指肠和结肠的吸收量次之,依次为338.5、318.8、228、201.5μg·h-1。结论 氧化苦参碱在小肠各肠段均无特异吸收,适合制成缓释制剂。

    Study on in vitro absorption of oxymatrine from rats’ intestine

    Abstract: Objectives To study the absorption rate at different segment of rats and to determine if specific absorption site existed and the feasibility of the extended release preparation. Methods: Everted gut sac was used to compare the absorption of oxymatrine in jejunum, ileum, duodenum and colon. The concentration of oxymatrine in the intestinal medium was determined by HPLC method. Results: The highest absorption extent of oxymatrine was observed in the jejunum and ileum and the lower absorption rate was observed in duodenum and colon, with 677, 637, 456 and 403μg·h-1, respectively. Conclusions: No significant difference in absorption rage between each segment of the rats’ intestine. It was possible to formulation oxymatrine to extented release dosage forms according to the absorption study.
