异常黑胆质成熟剂与清除剂对氧化损伤的淋巴细胞p53蛋白表达的影响

     摘要: 目的:探讨人淋巴细胞氧化损伤时p53基因表达的变化和异常黑胆质成熟剂与清除剂对其的影响,研究异常黑胆质成熟剂与清除剂抗氧化损伤的机理。方法:建立人淋巴细胞氧化损伤模型,电镜观察细胞变化,分别应用H2O2、异常黑胆质成熟剂与清除剂及两者联用处理, 以及Western blot检测p53蛋白表达变化。结果:淋巴细胞氧化损伤时, p53蛋白表达量明显增加;异常黑胆质成熟剂与清除剂同时对H2O2损伤的p53蛋白表达均有抑制作用。结论:异常黑胆质成熟剂与清除剂可能是通过清除氧自由基,影响凋亡基因p53的表达,从而有效地抑制氧自由基对人淋巴细胞的损伤。

    Influence of Abnormal Savda Munziq and Mushil on p53 protein and gene expression oxidized of human lymphocytes in vitro

    Abstract: Objective: To investigate the effect of Xinjiang Uighur medicine Savda Munziq and Mushil on the changes of p53 expression occur in the oxidized human lymphocytes. Methods: The experimental model of oxidized was made. Morphological analysis of oxidized lymphocytes by electron microscopy, and treated with H2O2 、Savda Munziq and Mushil in various groups. Meanwhile, the level of p53 protein was evaluated by Western blot. Results: The protein of p53 gene is increased in human lymphocytes with H2O2 treatment. Savda Munziq and Mushil can inhibit this effect. Conclusion: The p53 protein is increased in oxidized human lymphocytes. The mechanism of Xinjiang Uighur medicine Savda Munziq and Mushil can eliminate oxygen free radicals, and change the expression of p53 gene, so as to inhibit the damage of human lymphocytes caused by oxygen free radicals. ......
