AFLP结合SCAR技术在中药材道地性鉴别中的应用

     【摘要】 道地药材和非道地药材在形态、生药性状及化学成分等特征上具有高度相似性,因而传统的鉴别方法对道地药材的鉴别往往不易。本文首次将扩增片段长度多态性技术(AFLP,amplified fragment length polymorphism)应用到中药材道地性的鉴别中,绘制不同产地中药材的AFLP指纹图谱和遗传多样性,确定道地性产区药材的特异性差异带,从而区分道地药材。在此基础上,将AFLP标记转换成简单实用的SCAR(特征性片段扩增区域,sequence characterized amplified region)标记。将SCAR标记片段进行胶回收,PCR或克隆测序,然后根据序列特点设计特异性引物,通过简单的PCR,即可鉴别道地性产地中药材。在资源调查、鉴定研究的基础上,建立中药材道地性评价体系,为最终从分子生物学水平上建立中药材道地性的统一质量控制标准,提供简便、快速、准确的鉴别技术。
