两种介导物对人视网膜色素上皮细胞体外转移外源基因的效率

    摘 要:目的 比较两种介导物多阳离子脂质体(lipofec-tamine)及一种具有专利的脂类混合物(FuGENE6)的基因转移效率. 方法 体外培养人视网膜色素上皮细胞(RPE),并以携带β-半乳糖苷酶报道基因的多阳离子脂质体和脂类混合物转移人视网膜色素上皮细胞,以X-gal染色系统检测. 结果 分别以多阳离子脂质体及脂类混合物转移6h和表达48h后,在人视网膜色素上皮细胞中β-半乳糖苷酶的阳性表达比例为47%和6%. 结论 在体外能够由第二代多阳离子脂质体有效地将外源基因转移至人视网膜色素上皮细胞,脂类混合物仅获得较低的转染效率.

    Abstract:AIM To study the efficiency of gene transfer with polycationic liposomes(lipofectamine)or a proprietary blend of lipids(FuGENE6).METHODS Lipofectamine or Fu-GENE6carrying the beta-galactosidase report gene were transfered into the human retinal pigment epithelial(RPE)cells in vitro and examined following X-gal staining.RESULTS 47%and6%of beta-galactosidase expression in the RPE cells were positive at48hours post-transfer with lipofectamine and FuGENE6respectively.CONCLUSION The exogenous gene can be efficiently transfered onto the RPE cells by lipofectamine in vitro ......
