【Abstract 】 AIM: To elucidate the therapeutic effects of thymocytes microencapsulated with agarose transplanted into systemic lupus erythematosus (SLE) mice. METHODS: Thirtyfive BALB/c×C57BL/6 F1 mice were divided into 5 groups after their being injected with their parental lymphocytes suspended solutions. When the models were constructed successfully, group A accepted a therapy by physiological saline combined with hollow microencapsulations; Group B accepted a therapy by adrenaline combined with hollow micoencapsulations; Group C physiological saline and microencapsulated thymocytes; Group D adrenaline and microencapsulated thymocytes; Group E physiological saline and suspended thymocytes solution. Twentyfive days after the therapies, mice serum were detected by flow cytometry (FCM) for T cells ratio of CD4+/CD8+, Thymus were weighed with an electronic equiarmbalance, Direct immunofluorescence technique was used to test the intensities of fluorescence of mice kidney frozen sections, The fluorescence titres of immune complex deposited at mice glomerular basement membrane were tested by indirect immunofluorescence technique. RESULTS: ① There were not apparent fibrous tissues observed on the outmembrane of microencapsulations. ② The T cells CD4+/CD8+ ratio and the serum titres of antinuclear antibody (ANA) in group A were higher than that of group C (P<0.05); The wet weight of thymus of group C was higher than that of group A (P<0.05); On the other hand, the fluorescence intensity of mice kidney frozen sections in group C was weaker than that of group A (P<0.05). ③ The T cells CD4+/CD8+ ratio and the serum titres of ANA in group C were higher than that of group D (P<0.05); The wet weight of thymus of group D was higher than that of group C (P<0.05); On the other hand, the fluorescence intensity of mice kidney frozen sections in group D was weaker than that of group C (P<0.05). CONCLUTION: ① Agarose was a kind of solubilitywell biomaterial. ② Microencapsulated thymocytes made a significant therapeutic effect to SLE mice after their being transplanted into model animals’ abdomens. ③ Adrenaline therapy combined with microencapsulated thymocytes transplantation got a better result than anyone of the individual therapies. ......
