     Cloning and sequencing of human heparanase gene from hepatoma

    【Abstract】 AIM: To study the role of heparanase in the growth and metastasis of human hepatocelluar carcinoma (HCC), and to colone heparanase cDNA gene from HCC. METHODS: The cDNA fragment encoding heparanase was isolated by RTPCR method with the total RNA extracted from human hepatoma, linked into pGEMT easy vector, then amplified, cloned, finally the recombinant plasmids was identificated by Electrophoresis. RESULTS: The result of sequencing indicated that the sequences of our cDNA was identical to the corresponding parts of the mRNA for human heparanase in GenBank except 16 nucleotides, and with a homoloty of 0.991. CONCLUSION: Human heparanase cDNA gene is successfully cloned from HCC.
