Expression and significance of 5ª²HT3 receptors in the ovariectomy restraint stress rats colon

    ¡¾Abstract¡¿ AIM: To investigate the effects of ovarian hormone on the expression of 5ª²HT3 receptors in the restraint stressª²induced dysfunction rats colon. METHODS: Twentyª²four female SD rats were randomly divided into three groups: Sham operation; ovariectomy (OVX) and ovariectomy with estrogen (E2) and progesterone (P) replacement therapy (OVX+E2+P). The rats were given 1 h restraint stress 4 week after operation. The effect of restraint stressª²induced on fecal pellets and the time of glass pellets output were observed; and the expression of 5ª²HT3 receptors mRNA in the colon was studied by RTª²PCR. RESULTS: In contrast with the sham group and OVX+E2+P group, OVX group showed increase in the fecal pellets output and decrease in the time of glass pellets output (P<0.01); and the level of 5ª²HT3 receptors mRNA in ovariectomied rats colon was significantly increased, the expression of 5ª²HT3 receptors mRNA was decreased in the hormone replacement therapy (P<0.01). CONCLUSION: These results indicate that ovarian hormone plays an important role in regulation 5ª²HT3 receptors in irritable bowel syndrome. ......
