     Reverse effect of Chinese Drug GLM on K562/ADM and MCF/ADM cell

    【Abstract】 AIM: To investigate the effect of Chinese drug GLM (Gonglaomu) initial solution on K562/ADM leukemia resistant cell and MCF7/ADM breast cancer resistant cell line, in order to search new effective agents reversing MDR of tumor cell. METHODS: MTT (Mehylthazoltetrazolinum) assay and trypan blue antistaining test were used to determine the cytotoxic effect of ADM, when combined with GLM, on K562/ADM cell line and MCF7/ADM cell line. Rhodamin 123 fluorescent dye retention test was used to assess the effect of GLM on intracellular concentration. RESULTS: MTT assay showed that the inhibition rate of K562/ADM and MCF7/ADM cell in GLM used group was higher than that in GLM nonused group (P<0.01). Trypan blue assay showed that the survival rate of K562/ADM, MCF7/ADM in GLM used group (34.5% and 38.5%) was lower than that in GLM nonused group (68.2% and 77.4%, P<0.01). Reverse multiple of K562/ADM, MCF7/ADM cell was 6.28 and 5.78 respectively. Rh123 retention test suggested that GLM in combination with ADM could increase the intracellular ADM concentration. CONCLUSION: GLM could enhance the cytotoxic effect of ADM on drugresistant cell and increase intracellular drug concentration in a concentrationdependent fashion. ......
