Effects of clozapine, risperidone and haloperidol on plasma leptin in firstª²episode schizophrenic patients

    ¡¾Abstract¡¿ AIM: To explore the effects of clozapine, risperidone and haloperidol on plasma leptin in firstª²episode schizophrenic patients. METHODS: Ninetyª²five firstª²episode schizophrenics were randomly divided into three groups, which were treated with clozapine, risperidone and haloperidol, respectively. The plasma leptin concentration was measured with radioimmunoassay pre treatment and 6 weeks postª²treatment. RESULTS: ¢Ù Plasma leptin level of postª²treatment [(8.0¡À5.2) ¦Ìg/L vs (8.1¡À4.4) ¦Ìg/L, respectively] increased compared with that of preª²treatment [(6.4¡À4.9) ¦Ìg/L vs (7.1¡À3.9) ¦Ìg/L, respectively] in both clozapine group(P<0.01)and risperidone group(P<0.05), and significant difference of the increased value of plasma leptin[(1.6¡À1.3) ¦Ìg/L vs (0.3¡À1.1) ¦Ìg/L, respectively]was found after treatment between clozapine and haloperidol groups(P<0.05). ¢Ú Body mass [(60.3¡À11.7) kg vs (56.2¡À10.6)kg, respectively] after treatment increased compared with that before treatment [(57.9¡À11.5) kg vs (55.0¡À10.6) kg, respectively] in both clozapine group(P<0.01) and risperidone group(P<0.01)and significant difference of the body mass gain was found between clozapine group and risperidone, haloperidol groups respectively (P<0.05). ¢Û The increased values of plasma leptin at the 6th weekend were positively correlated to the body mass gain in both clozapine group(r = 0.808, P<0.01) and risperidone group (r = 0.757, P<0.01). CONCLUSION: Both clozapine and risperidone have significant effects on plasma leptin concentration and body mass, and clozapine has stronger effect than risperidone on body mass, while haloperidol has no obvious effect on plasma leptin and body mass. ......
