    摘要: 目的: 建立牛膝总甾酮含量测定的新方法。方法: 采用大孔吸附树脂提取分离牛膝总甾酮, 以50mg·mL - 1

    牛膝为苋科植物牛膝(A chy ranthes bid entata B I . )的干燥根,主产于河南省, 具有补肝肾、强筋骨、活血通经的作用。甾酮是牛

    膝有效成分, 主要有蜕皮甾酮, 牛膝甾酮, 红苋甾酮等[1 ]。文献报

    道甾酮的含量测定多采用高效液相色谱法[2, 3 ]

    , 关于牛膝总甾酮

    的含量测定方法, 尚未见报道, 本文用分光光度法测定牛膝总甾

    酮的含量, 现报道如下。

    1 仪器与材料

    供试药材购于重庆桐君阁医药公司, 经重庆市中药研究院傅

    善权副主任技师鉴定, 为怀牛膝(A . bid entata)的干燥根; 蜕皮甾

    酮对照品由本实验室自制; 大孔吸附树脂(天津制胶厂) ; 所用试

    剂均为分析纯; 水为重蒸水。安捷伦 7530G 分光光度计。

    2 方法与结果

    2. 1 对照品溶液的制备 精密称取干燥至恒重的蜕皮甾酮对照

    品, 用 100%甲醇溶解制成 5. 02 mg·m l

    - 1

    溶液, 作为对照品溶

    液。

    2. 2 样品溶液的制备 牛膝饮片适量, 分别加入 20, 15 倍量水,在90～ 100℃下水煮2 次, 2 h? 次。过滤收集牛膝提取液。提取液

    通过大孔吸附树脂柱, 洗脱液于 80℃烘干至恒重, 得到牛膝提取

    物。精密称取牛膝提取物适量, 用100%甲醇制成5 mg·m l

    的

    溶液, 作为供试样品溶液。
