    【Abstract】 AIM: To study the effect of extracellularregulated kinase signaling pathways on typeⅠ collagen synthesis and TGFβ1 expression stimulated by acetaldehyde in rat hepatic stellate cells (HSCs). METHODS: HSCs were isolated from liver of SpragueDawdey rats by in situ perfusion with pronase and collagenase and density gradient centrifugation with metrizamide and cultured. TypeⅠ collagen and transforming growth factor beta 1 mRNA expression in HSCs, stimulated by acetaldehyde predisposed in different doses of PD98059, were detected with ELISA and RTPCR. RESULTS: PD98059 with a concentration of 20 μmol/L suppressed typeⅠ collagen synthesis and TGFβ1 expression in HSCs stimulated by acetaldehyde (P>0.05), and the inhibitory effect of PD98059 at the concentration of 50 μmol/L and 100 μmol/L was significant (P<0.05 or P<0.01)with a dose dependant effect. The TGFβ1 expression and typeⅠ collagen synthesis had a positive correlation (r=0.93). CONCLUSION: Erk signaling pathways in HSCs stimulated by acetaldehyde play an important role in the regulation of Ⅰ collagen synthesis and the expression of TGFβ1 mRNA. The expression of TGFβ1 mRNA inhibited by PD98059 may trigger the decrease of typeⅠ collagen synthesis of HSCs stimulated by acetaldehyde. ......
