     Genetic polymorphism of 12 STR loci in the Han nationality in Jilin

    【Abstract】 AIM: To analyze the genetic polymorphism of 6 short tandem repeat (STR) double loci, namely, 12 STR loci and to obtain the corresponding genetic data. METHODS: DNA was extracted from the blood cells of 103 unrelated individuals of Han population in Jilin. Twelve loci were divided into 6 groups: group Ⅰ (TPOX+CSF1PO), group Ⅱ (D3S1358+D13S317), group Ⅲ (D5S818+D19S400), group Ⅳ (VWA+D12S391), group Ⅴ (D1S1656+FGA) and group Ⅵ (D8S1179+D19S253) and were amplified with PCR multiplexing method. The PCR products were analyzed by PAGE, followed by silver staining. The allele frequency distributions of the 12 STR loci were studied. The heterozygosity (H), discrimination power (DP), probability of exclusion (PE) and polymorphic information content (PIC) were calculated. RESULTS: The allele frequencies of the 12 loci were obtained. The genotype distributions of the loci were consistent with HardyWeinberg equilibrium and had higher heterozygosities and PIC. The H, DP, PE and PIC were 0.7511-0.8524, 0.8214-0.9580, 0.5167-0.6956 and 0.7100-0.8358 respectively. The accumulative DP was 0.9999999999992 and the accumulative PE was 0.999988. CONCLUSION: The 6 STR double loci have higher polymorphism and are satisfactory genetic markers. The data on the allele frequencies of these 12 STR loci can be used in individual identification, paternity testing and other population genetic researches. ......
