Effects of IFITM1 genes on proliferation and adhesion of colorectal carcinoma cell line SW480

    ¡¾Abstract¡¿ AIM: To investigate the effects of interferon induced transmembrane protein 1 (IFITM1) on the proliferation and heterotypic adhesion of SW480 cell line. METHODS: Recombinant vectors of pEGFPª²C3/IFITM1were constructed and were then transfected to the SW480 cell line. After being selected by G418, the subclone cell line was obtained. Laser confocal microscopy (LCSM ) was used to examine green fluorescence protein (GFPª²IFITM1) for the gene expression position in cell line. The proliferation of cell line was detected by the cell growth curve and adhesion of cell line was evaluated by cell adhesion assay. RESULTS: The sequence of recombinant vectors of pEGFPª²C3/IFITM1 was consistent with the IFITM1 sequence. After SW480 cells being transfected by eukaryotic expression vector of pEGFPª²C3/IFITM1, the subclone cell line IFITM1/SW480 was obtained by persistent G418 selection. The findings of LCSM showed that GFPª²IFITM1 protein was located in IFITM1 cell membranes, suggesting correct expression of IFITM1 genes after transfection. The cell growth curve showed that the proliferation of IFITM1/SW480 was reduced. The adhesive cell numbers of IFITM1/SW480 were(3.93¡À0.08)¡Á103 and (6.10¡À0.21)¡Á103 at 30 min and 60 min, respectively, dramatically lower than those in the control groups of pEGFPª²C3/SW480 [(4.61¡À0.25) ¡Á103, (7.17¡À0.22) ¡Á103 at 30 min and 60 min, respectively (P<0.05)]. CONCLUSION: The IFITM1 can inhibit the proliferation and heterotypic adhesion of SW480 cell line in vitro. ......
