     Study on biological properties of mouse embryonic stem cells modified by pTracer/mIFNγ

    【Abstract】 AIM: To study the characteristics of embryonic stem (ES) cells transfected with pTracer/mIFNγ. METHODS: The gene pTracer/mIFNγ including IFNγand green fluorescent protein (GFP) was transfected to obtain ES/ pTracer/mIFNγ cells by electroporation. RTPCR and ELISA were used to detect IFNγ expression at transcription and protein level respectively and green fluorescence was detected with fluorescence microscope to prove the GFP expression. Colony formation rates for ES/pTracer/mIFNγ cells and ES cell were conducted respectively. ES/pTracer/mIFNγ cells were induced to differentiate into neural precursor cells (NPCs) in serumfree selective medium and nestin immunoreactive was identified by immunocytochemistry. RESULTS: Green fluorescence was observed in ES/pTracer/mIFNγ cells and IFNγ concentration was (44.25±12.23)μg/L 72 h after gene transfection. ES/pTracer/mIFNγ cells were positive to alkaline phosphatase. The cell colony formation rates of ES cells and ES/pTracer/mIFNγ cells were 73.66% and 70.89%, respectively (P>0.05). ES/pTracer/mIFNγ cells differentiated into NPCs, which had potency to differentiate into all kinds of neural cells. CONCLUSION: The cells modified with IFNγgene may not influence the proliferation, differentiation and the pluripotentiality of ES cells. ......
