Development and primary application of immunogold fastª²detection kit for human cytomegalovirus

    ¡¾Abstract¡¿ AIM: To develop a cheap and easyª²toª²use kit based on dot immunogold filtration assay for analysis of antibody of the human cytomlegalovirus in serum. METHODS: With prepared GICA apparatus and colloidal gold which were coupled with SPA, the goldª²SPA conjugate reacted with human IgG in serum and migrated along the nitrocellose strip on which the branched multiple antigenic peptide of human cytomlegalovirus was immobilized. If the serum contained antiª²HCMV IgG, there would be a red line on the strip where the branched multiple antigenic peptide of human cytomlegalovirus were immobilized. RESULTS: A total of 120 sera were detected for antiª²HCMV IgG by GICA and ELISA. The sensitivity and specificity of GICA were 97.9% and 87.5%. The corresponding rate of both tests was 95.8%. CONCLUSION: GICA is a sensitive, specific, simple and rapid assay for detecting antiª²HCMVIgG in sera. ......
