    胰岛素样生长因子2Ⅰ对早期胚胎发育终止的影响

    【摘要】 目的:探讨胰岛素样生长因子2Ⅰ( IGF2I)在早期胚胎发育终止绒毛中的表达

    以及母体血中 IGF2I水平改变与胚胎发育终止的关系。方法:采用免疫组化法(SP法)检

    测早期胚胎发育终止绒毛中 IGF2I蛋白的表达 ,采用原位杂交法( ISH)检测 IGF2I mRNA的

    表达 ,以正常妊娠为对照组。采用酶联免疫吸附试验(ELISA)检测正常妊娠及早期胚胎发

    育终止孕妇血中 IGF2I水平的变化。结果:正常妊娠绒毛中 IGF2I、 IGF2I mRNA 的表达均

    明显高于胚胎发育终止组( P < 0. 01) ;正常妊娠组母血中 IGF2I水平显著高于胚胎发育终

    止组( P < 0. 01) 。结论: IGF2I在绒毛组织中的表达及母血中的水平与胚胎发育密切相关 ,其含量减少可能是导致胚胎死亡的重要原因之一。

    【Abstract】 Objective :To investigate the relationship of villus expression and serum level of

    insulin 2like growth factor2I ( IGF2I) and embryogenesis termination of early pregnancy. Methods :

    Expression of IGF 2I was measured by immunohistochemistry. The expression of IGF 2I mRNA was de2

    tected by in situ hybridization ( ISH) . Serum IGF2I level was determined by enzyme2linked im2

    munoabsorbent assay (ELISA) . Results :Both the expressions of IGF 2I protein and mRNA were lower

    in the abnormal villus of embryogenesis termination than that of normal villus ( P < 0. 01) . Serum

    level of IGF2I in normal pregnancy women was much higher than that of abnormal pregnancy women.

    Conclusion :IGF2I may play an important role in the regulation of fetal growth.Decrease of IGF 2I ei2

    ther in villus or in serum may be one of the key factors contributing to the embryogenesis termination

    of early pregnancy.

    胚胎在宫内生长发育是一个多因素调节的过

    程 ,研究表明 ,多种细胞因子在正常妊娠维持过程中

    起着很重要的调节作用 ,其中胰岛素样生长因子

    (insulin2like growth factors , IGFs) 是维持正常妊娠和

    胎儿生长发育的重要调节因子之一 ,妊娠过程中母

    血循环中的 IGFs 浓度逐渐增加 ,产后含量迅速降

    低。因此 ,推测 IGFs 在早期胚胎发育过程中可能

    起着重要作用。最近 ,Servoss 等1

    证实 , IGF2I具有

    调节胎儿生长发育的作用。其具体作用途径和作用

    机制需要进一步探讨。我们通过检测正常胚胎发育

    孕妇及胚胎发育终止孕妇血中的 IGF2I水平和绒毛

    中的 IGF2I表达的变化 ,初步探讨了 IGF2I在早期胚

    胎发育中的作用。

    1 材料和方法

    1. 1 材料 (1)待检标本:以 2002 年 4 月至 2003 年 2 月在

    我院妇产科门诊经B超证实胚胎发育终止而行清宫术 48 例
