复方中药“克瘤宁”对人食管癌的抗癌作用及其机理研究

     摘 要:目的:探讨复方中药“克瘤宁”对人食管癌的抗癌作用及其机理。方法:① 应用光镜、透射电镜,观察复方中药“克瘤宁”诱导人食管癌细胞株EC109细胞凋亡的作用;② 用“克瘤宁”对移植人食管癌8d后的裸鼠进行治疗,实验共分五组:“克瘤宁”高浓度组(2.4g/kg)、中浓度组(1.2g/kg)、低浓度组(0.6g/kg)、阳性对照组(平阳霉素15mg/kg)和阴性对照组(生理盐水)。“克瘤宁”灌胃给药,每天一次,连续给药36次,平阳霉素腹腔注射给药,每3d一次,共给药12次。实验重复二次。结果:① 经“克瘤宁”处理的EC109细胞出现典型的细胞凋亡特征;② 克瘤宁高、中、低三剂量在裸鼠体内对食管癌的抑瘤率分别为:66.36%~76.85%(P<0.01)、54.21%~55.56%(P<0.01)、38.32%~39.81%(P<0.01),阳性对照平阳霉素在裸鼠体内对食管癌的抑瘤率为51.40%~56.48%(P<0.01)。结论:① “克瘤宁”有诱导人食管癌细胞产生凋亡的作用,这可能是“克瘤宁”抗食管癌作用的重要机理;② 在裸鼠可耐受的剂量下,“克瘤宁”对裸鼠移植的人食管癌有明显抗癌作用,且随着剂量增加,其抗癌疗效增加。

    Antitumor Effect of Zhenhuang Capsule on Human Esophageal Carcinoma and its Mechanisms

    Abstract: Objective: To investigate the antitumor effect of Zhenhuang capsule(ZHC) on human esophageal carcinoma and research its mechanisms. Methods: ①Apoptosis induced by ZHC in human esophageal carcinoma cell line EC109 was investigated by light microscopy, transmission electron microscopy combined with flow cytometry analysis technique. ② A perfusing test with ZHC was conducted on nude mice 8 days after the human esophageal carcinoma transplantation. The animals were divided into 5 groups: high ZHC dose group (2.4g/kg, BW), medium ZHC dose group (1.2g/kg), low ZHC dose group (0.6g/kg), positive control group of Pingyangmycin (15mg/kg) and saline control group. ZHC was perfused into stomach, once daily, with 36 times consitituting one course. Pingyangmycin was injected intraperitaneously, once every three days, 12 times consitituting one course. The test was repeated 2 times.Results: ①After exposure to ZHC, EC109 cells manifested typical apoptotic features. ②The tumor-inhibiting rates of the high, medium and low dose ZHC groups ranged from 66.36% to 76.85%(p<0.01), 54.21% to 55.56%(p<0.01) and 38.32% to 39.81%(p<0.01), respectively. The tumor-inhibiting rate of the Pingyangmycin group ranged from 51.40% to 56.48%(p<0.01). Conclusion: ①ZHC induces apoptosis of esophageal carcinoma cells and ZHC induced apoptotsis possibly the primary mechanism of ZHC for treatment of esophageal carcinoma. ② ZHC has significant antitumor effect on human esophageal carcinoma transplanted into nude mice receiving tolerance doses. The effect increase with elevation of dose. ......
