【Abstract】 AIM: To investigate the protective effects of Interleukin10 (IL10) on ischemia/reperfusion (I/R)induced lung injury following rat lung transplantation and to explore its mechanisms. METHODS: Thirtysix SD rats were randomly divided into 3 groups with 12 rats in each group: sham operation group (control), lung transplantation group and IL10 treatment group. IL10 was administered to the recipient rats through intratrachea way. The wet/dry ratio of lung, lung permeability index and neutrophil percentage in bronchoalveolar lavage fluid (BALF) were detected. The serum level of tumor necrosis factor α(TNFα)was detected by enzymelinked immunosorbent assay (ELISA). RESULTS: The wet/dry ratio of lung, lung permeability index and neutrophils percentage in BALF of I/R group were (657±089),(2349±513)×10-4 and (4984±653)%, significantly higher than those of the control group(P﹤001). The administration of IL10 to recipient rats before the operation markedly reduced the indexes above to (423±058), (1596±304)×10-4 and (2033±426)%, respectively(P﹤001). The serum level of TNFα(μg/L)increased to (086±011) after lung transplantation, markedly higher than that of control group, however, the IL10 treatment could reduce its level significantly to [(039±005),P﹤001]. CONCLUSION: IL10 protects the lung function against I/R injury after transplantation, which is related to its inhibition of TNFα release and activation of neutrophils. ......
