三萜类化合物(双灵固本散)抑制肿瘤细胞的实验研究

     【摘要】 目的 研究灵芝主要成分三萜类化合物Ganodermenonol对胃癌和肝癌细胞生长的抑制作用。方法 (1)运用硅胶柱色分离法,从双灵固本散的氯仿提取物中分离三萜化合物Ganodermenonol。(2)采用经典化疗药物顺氯氨铂做对照,系统地研究双灵固本散的不同溶剂提取物对胃癌细胞和肝癌细胞的抑制作用;同时进行两种癌细胞抑制生长效果对照。(3)实验分析双灵固本散中氯仿提取物及其有效成分的含量测定。结果 (1)在双灵固本散药物中,每个溶剂的提取物都有一定的抗癌细胞作用,抗癌细胞作用较好的活性物质主要集中在非极性提取物中,其中三萜类化合物是其活性最强的成分之一,具有较高含量的高效抗癌成分Ganodermenonol在微克级即表现出明显抑癌细胞作用,是该药的最大特色。(2)双灵固本散的氯仿提取物及乙酸乙酯提取物对肝癌细胞BEL-7402,在浓度为0.125mg/ml时抑瘤率超过50%;三萜类化合物Ganodermenonol对肝癌细胞BEL-7402,在1.5μg/ml的剂量时抑瘤率超过50%,高达64%。氯仿提取物及乙酸乙酯提取物对胃癌细胞MGC-803在浓度为0.125mg/ml时抑瘤率高达83.1%。(3)双灵固本散中氯仿提取物占7.1%,乙酸乙酯占5.3%,Ganodermenonol占0.37%(w/w)。结论 双灵固本散所含三萜类化合物Ganodermenonol在微克级有明显抑制肿瘤细胞生长作用。

    Research on the tumor inhibiting action of triterpenoid compound (Sunrecome)

    【Abstract】 Objective Ganoderma Lucidum is a traditional Chinese medicine of Fuzheng Guben. Experiments show that ganodermenonol, one of triterpenoid compounds which is the main component of Ganoderma Lucidum, has an inhibiting action to the growth of gastric carcinoma and liver cancer.Methods (1)Separation method of silica gel column chromatography, ie, the triterpenoid compound ganodermenonol separated from the chloroform extraction of Sunrecome has a strong action to inhibit the activity of cancer cells. (2) Using cisdiaminedichlorop , the classic drug in chemical therapy, as a contrast, we tried to find out whether the essence extracted from different solutions of Sunrecome has the same inhibiting action to gastric carcinoma and liver cancer. Meanwhile, we compared the inhibiting effect on the two different kind of cancers.(3) We estimated the exact quantity of the chloroform extraction of Sunrecome and its effective component.Results (1)Each one of the essence extracted from different solutions of Sunrecome has a certain effect on inhibiting the proliferation of cancer cell. While the more effective can be found in the non-polarity solution of Sunrecome, and one of the most active component is a triterpenoid compound called ganodermenonol, this compound can show its tumor inhibiting action at microgram level , constitutes the most attracting feature of this drug.(2) The chloroform extraction and the ethyl acetate extraction of Sunrecome can inhibit over 50% of the liver cancer cell BEL-7402 at the concentration of 0.125mg/ml; The triterpenoid compound ganodermenonol can inhibit as high as 64% of the liver cancer cell BEL-7402 at the dose of 1.5 ug/ml. As for the stomach cancer cell MGC-803, the chloroform extraction and the ethyl acetate extraction of Sunrecome can inhibit 83.1% of the tumor at the concentration of 0.125mg/ml.(3) The chloroform extraction makes up 7.1% of Sunrecome, and 5.3% for ethyl acetate extraction , 0.37% for ganodermenonol (w/w).Conclusion Ganodermenonol contained in sunrecome has an obvious effect on inhibiting the growth of tumor at the level of microgram. ......
