摘要:目的 了解更年期大鼠脾细胞凋亡与辅助性T细胞1(Thelper l, Th1)和辅助性T细胞2(Thelper 2, Th2)型细胞因子的增龄性改变。方法 应用Annexin VFITC/PI双染色流式细胞仪测定脾细胞凋亡率; ELISA法测血清γ干扰素(interferonγ, IFNγ)、白介素4(interleukin4, IL4)水平。结果 更年期组大鼠脾细胞凋亡率(10.3% vs. 7.7%, P<0.05)、血清IFNγ质量浓度明显高于年轻组大鼠(23.80ng/L vs. 10.92ng/L,P<0.01);更年期组大鼠与年轻组相比血清IL4质量浓度有下降趋势,但差别无显著意义(0.1296ng/L vs. 0.3094ng/L, P>0.05);更年期组血清IFNγ/IL4比值明显高于年轻组(183.64 vs. 35.29,P<0.01),Th1/Th2平衡朝Th1漂移。结论 大鼠衰老过程中Th1/Th2平衡朝Th1漂移,脾细胞凋亡增加与IFNγ水平增高有关。关键词:更年期;脾细胞;凋亡;Th1;Th2
The changes of Th1/Th2 balance and apoptosis in splenocyte of menopausal rats

ABSTRACT: Objective To investigate the relation between the apoptosis in splenocyte and the changes of Th1/Th2 balance in serum of menopausal rats. Methods Twenty female SD rats were divided into two groups: menopausal group(n=10;15 months), young group (n=10; 5.5 months). The apoptosis in splenocyte of rats was measured by Annexin VFITC/PI flow cytometric assay. The levels of IFNγ and IL4 in serum were measured by ELISA. Results The percentages of apoptosis cells in splenocyte of menopausal rats were significantly higher than that of young rats. The serum IFNγ level of menopausal rats was significantly higher than that of young rats(23.80ng/L vs. 10.92ng/L, P<0.01). The level of IL4 was similar in menopausal and young groups. Conclusion The increasing of apoptosis in splenocyte of menopausal rats is related to the increasing of IFNγ levels with aging. ......
