【Abstract】AIM: To investigate the inhibition of lung metastasis of melanoma by a tumor vaccine of B16 melanoma cells transfected with murine granulocytemacrophage colonystimulating factor (mGMCSF). METHODS: The murine melanoma cell line B16 was transfected with the recombinant plasmid containing mGMCSF. A cell line stably expressing mGMCSF was obtained by G418 selection. The Balb/c mice were administered i.v. with B16 cells transfected with or without mGMCSF respectively and the lung metastasis was examined 2 weeks later. RESULTS: The recombinant cell line B16TR was established after mGMCSF tranfection and the expression of mGMCSF was confirmed by RTPCR. The number of lung metastatic lesions of mice treated with B16TR was significantly fewer than that of mice treated with B16 (n=5, 125.8±34.3 vs 27.8±24.0, P<0.01). CONCLUSION: The mGMCSF transfection can significantly suppress the lung metastasis of melanoma in mice. The GMCSF can be a promising adjuvant in the immunotherapy of melanoma. ......
