     Osteogenesis of nacre/PDLLA artificial bone combined with osteoblasts in repairing rabbit radius segmental defect

    【Abstract】 AIM: To survey the osteogeneic effect of Nacre/PDLLA (N/P) artificial bone combined with allogeneic osteoblasts in repairing rabbit radius segmental defect and to explore its application as scaffold material of bone tissue engineering.METHODS: The allogeneic osteoblasts,seeded onto N/P artificial bone,were cocultured in vitro for one week and then were implanted into the rabbit radii segmental defect sites of the New Zealand white rabbits.The defects of 15 mm long were produced on the bilateral radii of 18 adult rabbits by surgery.The PDLLA alone artificial bone combined with allograft osteoblasts and the N/P artificial bone without osteoblasts served as control.The repairing effect was evaluated at 4,8 and 12 weeks postoperation respectively by gross observation,radiograph examination and histological examination.RESULTS: In experimental group,with N/P artificial bone,masses of lamellar bone formed 8 weeks postoperation,and the radii segmental defects were fully bridged by the new bone tissue with sequence bone cortex and partly connected medullary cavities 12 weeks postoperation,which was confirmed by Xray and histological examination.Part of the N/P artificial bone became powder through degradation and it was evident around the new bone tissue.But in control group with PDLLA artificial bone combined with osteoblasts,less new bone mass formed.In PDLLA artificial bone materials,less lamellar bone and more osteoid tissue formed 8 weeks postoperation.Bulks of PDLLA artificial bone were degraded.After 12 weeks,the bone defects were bridged by sequence and narrow new bone tissue,but medullary cavities were blurry.PDLLA artificial bone completely degraded. In another control group with N/P artificial bone without osteoblasts,masses of new bone tissue formed at the joint of material and host bone,but less new bone formed in materials after 12 weeks.In inducing new bone formation,the artificial bone with osteoblasts was more effective than the material without osteoblasts.CONCLUSION: N/P artificial bone combined with allogeneic osteoblasts has better osteogenesis than PDLLA artificial bone with osteoblasts and N/P artificial bone without osteoblasts,which can effectively repair rabbit radius criticalsize defect.N/P artificial bone can be used as an optimal scaffold material of bone tissue engineering. ......
