【Abstract】 AIM: To investigate the effect of hypoxia on transforming growth factor3β(TGF3β) expression in trophoblasts and its regulation by hypoxiainducible factor1 (HIF1). METHODS: The BeWo cells,derived from trophoblasts,were divided into four groups to be cultured: control group,hypoxic group,antisense group and sense group.The cells in antisense group and sense group were incubated under hypoxic condition after cultured with HIF1α oligonucleotide under normal conditions for 6 h.After 48 h,the cells were harvested.The mRNA expression of HIF1α and TGF3β was evaluated by real time PCR and the protein expression of HIF1α and TGF3β was measured by Western blot.RESULTS: Compared with those in the control group,both the mRNA expression and the protein expression of HIF1α and TGF3β increased in hypoxic group.Compared with the sense group,the mRNA expression and the protein expression of HIF1α and TGF3β decreased in antisense group.CONCLUSION: The hypoxia can induce the expression of TGF3β in trophoblasts and the inducible effect of hypoxia on TGF3β expression may be mediated by HIF1. ......
