     Effects of hypoxia on transforming growth factor3β expression and its regulation in trophoblast

    【Abstract】 AIM: To investigate the effect of hypoxia on transforming growth factor3β(TGF3β) expression in trophoblasts and its regulation by hypoxiainducible factor1 (HIF1). METHODS: The BeWo cells,derived from trophoblasts,were divided into four groups to be cultured: control group,hypoxic group,antisense group and sense group.The cells in antisense group and sense group were incubated under hypoxic condition after cultured with HIF1α oligonucleotide under normal conditions for 6 h.After 48 h,the cells were harvested.The mRNA expression of HIF1α and TGF3β was evaluated by real time PCR and the protein expression of HIF1α and TGF3β was measured by Western blot.RESULTS: Compared with those in the control group,both the mRNA expression and the protein expression of HIF1α and TGF3β increased in hypoxic group.Compared with the sense group,the mRNA expression and the protein expression of HIF1α and TGF3β decreased in antisense group.CONCLUSION: The hypoxia can induce the expression of TGF3β in trophoblasts and the inducible effect of hypoxia on TGF3β expression may be mediated by HIF1. ......
