5氮杂胞苷对HT29细胞DLC.1基因表达和细胞增殖活性的影响

     [摘要]目的 探讨去甲基化药物5氮杂胞苷对肠癌细胞系HT29的DLC.1基因表达和细胞增殖活性的影响。 方法 使用浓度为5μmol·L-1 和10μmol·L-1 的去甲基化药物5氮杂胞苷处理HT29细胞,通过MTT实验观察细胞增殖活性的差异,RT.PCR技术检测DLC.1mRNA表达强度的变化。 结果 经药物处理后,HT29细胞的增殖活性受到抑制,DLC.1mR-NA的表达明显增强。 结论 HT29细胞DLC.1mRNA的表达下调可能是由于启动子区域高甲基化所致,DLC.1基因具有抑制肿瘤细胞生长的功能,其可能参与了结肠癌的发病机制。

    Abstract:Objective To explore the effect of5.Aza.CdR on gene DLC.1expression and cell proliferation in human colorectal cancer cell line HT29.Methods The colorectal cancer cell line HT29was treated with5μmol·L-1 and10μmol·L-1 5.Aza.CdR.The activation difference in cell growth was observed by MTT assay,and the changes of the DLC.1mRNA expression were detected by RT.PCR.Results Compared with the control,the cell growth was reduced after treatment,and the expression of the DLC.1mRNA was increased.Conclusion The ex-pression of DLC.1mRNA in the cell line HT29is downregulated resulting from the hypermethylation of CpG islands within the promoter regions ......
