防治母儿血型不合的抗溶冲剂的急性毒性试验及致畸敏感期毒性试验*

     【摘要】 目的 研究用于防治母儿血型不合的抗溶冲剂的用药安全性问题。方法 对抗溶冲剂进行了小鼠急性毒性试验,经预试实验无法测出其小鼠灌胃给药的LD50,故进行了小鼠灌胃该药,无明显毒性反应,最大耐受量的测定;应用Wistar大鼠对抗溶冲剂进行了致畸敏感期毒性实验。结果 ①小鼠灌胃给药最大耐受量为492g生药/kg,相当于人临床用量的240倍,无明显毒性反应;②该药对大鼠生殖能力、胎鼠生长发育(外观、内脏、骨骼)均无不良影响。结论 抗溶冲剂毒性很低,未发现胚胎毒性和致畸性,可用于孕期。

    Experimental observation of Kangrongchongji on acute toxicity test and the toxic influence on the fetus malformation sensitiveity state

    【Abstract】 Objective To observe the safety of Kangrongchongji on preventing the disorder of incompatable blood group between mother and fetus. Methods 20 mice , half male and half female,were fed three times continusly with Kangrongchongji solution after 12 hour's fast. Each feeding interval was five hours and the dosage was 0.4 mg/10 g.The fur color, weitght,appetite,bowel movement, activity mental state were observed after seven days feeding.? Wstar pregnant rats were randomly divied into nomal group, small dosage Kangrongchongji group, great dosage Kangrongchongji group and Vitamin AD group. Each pregnant rat was fed in the seventh day of pregnant and fed for ten days continuosly. In the ninteen day of pregnant, the whole pregnant rats were killed. The amount of fetus rat, the apperence, skeleton and viscera of the fetus rats were observed. Results The fur color, weitght,appetite,bowel movement, activity mental state were normal and no dead experimental mouce was observed.Compared with normal group,there were no significant difference of fetus rats in the small and great dosage Kangrongchongji groups in length, body weitht and plaenta weight(P>0.05).There were no malformal fetus rat in Kangrongchongji group and the skeleton was normal in the whole groups. Conclusions Kangrongchongji has low toxicity without malfromation effection. ......
