摘要:目的 研究TCR Vβ7.1基因在恢复肝癌病人细胞毒性T细胞(CTL)功能方面的作用。方法 以RTPCR方法扩增TCR Vβ7.1基因并构建Vβ7.1-pLXSN表达载体。重组体以脂质体转染外周血单个核细胞(PBLs),流式细胞仪检测转染效率,DNA Ladder实验和透射电镜观察转染后T细胞诱导肿瘤细胞凋亡情况。结果 TCR Vβ7.1基因转染PBLs后可有效表达并诱导肿瘤细胞凋亡。结论 该方法为T细胞介导的肿瘤基因治疗提供了有效途径。关键词:TCR Vβ7.1;转染;表达;凋亡
Expression and antihepatoma effect of TCR Vβ7.1 after being transfected into PBLs

Abstract: To study the effects of TCR Vβ7.1 gene in recovering the functions of cytotoxic T lymphocytes(CTL)in hepatoma, we amplified TCR Vβ7.1 gene by RTPCR and constructed expressive vector Vβ7.1 pLXSN. The recombinant was transfected into peripheral blood lymphocytes (PBLs) by Lipofectin Reagent. Transfection efficiencies of the TCR Vβexpressive vectors were tested by flow cytometric analysis. We also measured tumor cells apoptosis by DNA Ladder Assays and transmission electron microscopy. Our results show that TCR Vβ7.1transfectedPBLs can induce the apoptosis of tumor cells. TCR Vβ7.1 based T cell provides a useful approach to study specific aspects of immunetumor cells interactions and T cell mediated gene therapy. ......
