高效液相色谱法测定人血浆中全反式维甲酸血药浓度

     摘要:目的 建立高效液相色谱法测定人血浆中全反式维甲酸浓度。方法 色谱柱为Supelcosil LC18DB柱(5 μm,4.6 mm×250 mm);流动相为甲醇水冰乙酸(体积比82∶17∶1);柱温为室温;流速1.5 mL·min-1;检测波长为340 nm。结果 全反式维甲酸血药质量浓度在10~1000 ng·mL-1范围内,与峰面积比有良好的线性关系(r=0.9997),最低检测浓度为2.5 ng·mL-1。平均方法回收率为(94.5±4.7)%(n=15)。日内RSD≤4.1%,日间RSD≤6.2%。结论 本方法简单、快速、灵敏、重现性好,适用于全反式维甲酸临床血药浓度监测及人体药代动力学研究。

    Determination of alltrans retinoic acid in human plasma with HPLC

    Abstract: Objective To establish a HPLC method for determination of alltrans retinoic acid in human plasma. Methods A Supelcosil LC18DB column (5 μm,4.6 mm×250 mm) was adopted. Mobile phase was a mixture of methanol,water and glacial acetic acid(82∶17∶1). The flow rate was 1.5 mL·min-1. The UV detection wave was 340 nm. ResultsThe assay procedure was shown to produce linear calibration curves over the range of 10 ng·mL-1 to 1000 ng·mL-1 of alltrans retinoic acid in plasma (r=0.9997). Within the range,the recovery rate was (94.5±4.7)%,and the intraday RSD and interday was less than 4.1% and 6.2%,respectively. The detection limit of all trans retinoic acid was 2.5 ng·mL-1. Conclusions This method is repeatable,convenient and sensitive for plasma concentration monitoring and clinical pharmacokinetics study of alltrans retinoic acid. ......
