益气化瘀方对腰神经根损伤大鼠神经细胞黏附分子的作用

[摘要] 目的:探讨益气化瘀方对大鼠神经再生修复过程中神经肌肉接头部神经细胞黏附分子(neural cell adhesion molecule, NCAM)的作用。方法:大鼠L5神经根损伤后,予以益气化瘀方分别治疗10、20和30 d。采用免疫组织化学和激光共聚焦扫描显微镜技术,观察大鼠比目鱼肌神经肌肉接头部NCAM的分布变化。采用α金环蛇毒萤光结合剂显示乙酰胆碱受体(acetylcholine receptor, AChR),NIH图像分析系统定量测定NCAM与AChR的重叠面积。结果:益气化瘀方组NCAM在神经肌肉接头部的聚集、出芽、延伸以及与AChR的重叠面积均明显优于模型组。结论:NCAM的表达与AChR的神经支配情况密切相关。益气化瘀方可促进神经的再生修复。[关键词] 神经肌肉接头; 神经再生; 益气化瘀方; 神经细胞黏附分子
Effects of Yiqi Huayu Recipe on neural cell adhesion molecule in rats with lumbar nerve root compression

ABSTRACT Objective: To study the effects of Yiqi Huayu Recipe on neural cell adhesion molecule (NCAM) in neuromuscular junctions during nerve regeneration in rats with lumbar nerve root compression. Methods: The rats with lumbar nerve root compression were given Yiqi Huayu Recipe for 10, 20 and 30 days respectively. The distribution of NCAM in neuromuscular junctions of soleus muscle in rats was examined with immunohistochemical method and confocal laser scanning microscopy technique. The acetylcholine receptor (AChR) was visualized with fluoresceinconjugated αbungarotoxin (αBTX). The overlap areas of NCAM and AChR sites were measured with NIH image technique. Results: The aggregates, sprouts and extensions of NCAM in the neuromuscular junctions and the overlap areas of NCAM and AChR sites in the Yiqi Huayu Recipetreated group were all better improved than those in the untreated group. Conclusion: The expression of NCAM is regulated according to the state of innervation for muscles. Yiqi Huayu Recipe may accelerate this nerve regeneration process. ......
