摘 要 采用超临界CO2萃取技术对牡丹皮的有效部位进行萃取,并对其萃取物进行了鉴别及含量测定等方面的研究。采用薄层色谱法(TLC),以ψ(石油醚∶苯∶醋酸乙酯)=16∶2∶2为展开剂,ρ=5% FeCl3乙醇溶液为显色剂对牡丹皮超临界CO2萃取物中的丹皮酚进行鉴别;并建立以正十八烷为内标,采用SE-30(30 m×0.25 mm×0.25μm)石英毛细管色谱柱;进样口温度210 ℃,检测器温度300 ℃的气相色谱测定丹皮酚含量的方法。丹皮酚在ρ=99~693μg/mL浓度范围内,峰面积与内标峰面积之比值与浓度呈良好的线性关系(r=0.9999);平均回收率为97.12%,RSD=1.23%(n=6)。方法准确、简便、灵敏、快速。关键词 超临界CO2萃取技术;牡丹皮;丹皮酚;GC
Study on quality standard of the extracts in Paeonia suffruticosa Andr. by supercriticalCO2 fluid extraction (SFE)

Abstract The chemical substrates of the curative effect in Paeonia suffruticosa Andr. was extracted by supercritical- CO2 fluid extraction and the quality standard was studied. Paeonol in extract substrates of Paeonia suffruticosa Andr. was identified by TLC using a mobile phase of petroleum ether,benzene and ethyl acetate (16∶2∶2) and the color developing reagent of 5% Fecl3 ethanol. The content of paeonol was determined by capillary gas chromatography with n-octadecane as internal standard. The injector and FID detector temperatures were 210 ℃ and 300 ℃ respectively. The calibration curve was linear in the range of ρ=99~693μg/mL,r=09999. The average recovery was 97.12% (RSD=1.23%,n=6). This determination method was accurate,simple,sensitive and rapid. ......
