牡丹皮超临界CO2萃取物质量标准研究

     摘 要 采用超临界CO2萃取技术对牡丹皮的有效部位进行萃取,并对其萃取物进行了鉴别及含量测定等方面的研究。采用薄层色谱法(TLC),以ψ(石油醚∶苯∶醋酸乙酯)=16∶2∶2为展开剂,ρ=5% FeCl3乙醇溶液为显色剂对牡丹皮超临界CO2萃取物中的丹皮酚进行鉴别;并建立以正十八烷为内标,采用SE-30(30 m×0.25 mm×0.25μm)石英毛细管色谱柱;进样口温度210 ℃,检测器温度300 ℃的气相色谱测定丹皮酚含量的方法。丹皮酚在ρ=99~693μg/mL浓度范围内,峰面积与内标峰面积之比值与浓度呈良好的线性关系(r=0.9999);平均回收率为97.12%,RSD=1.23%(n=6)。方法准确、简便、灵敏、快速。

    Study on quality standard of the extracts in Paeonia suffruticosa Andr. by supercriticalCO2 fluid extraction (SFE)

    Abstract The chemical substrates of the curative effect in Paeonia suffruticosa Andr. was extracted by supercritical- CO2 fluid extraction and the quality standard was studied. Paeonol in extract substrates of Paeonia suffruticosa Andr. was identified by TLC using a mobile phase of petroleum ether,benzene and ethyl acetate (16∶2∶2) and the color developing reagent of 5% Fecl3 ethanol. The content of paeonol was determined by capillary gas chromatography with n-octadecane as internal standard. The injector and FID detector temperatures were 210 ℃ and 300 ℃ respectively. The calibration curve was linear in the range of ρ=99~693μg/mL,r=09999. The average recovery was 97.12% (RSD=1.23%,n=6). This determination method was accurate,simple,sensitive and rapid. ......
