hTCR Vβ 8.4基因修饰T细胞对SCID荷瘤小鼠的抑瘤作用研究

     摘 要 目的:探讨经hTCR Vβ8.4基因修饰的人外周血淋巴细胞(PBMC)转输给SCID小鼠的抑瘤作用。方法: 建立荷人肝癌细胞BEL-7402的SCID小鼠模型;用脂质体包裹hTCR Vβ8.4基因并转染健康人PBMC,将此PBMC回输入SCID小鼠体内,通过检测瘤重、肿瘤体积大小,并计算肿瘤抑制率来观察hTCR Vβ8.4基因的抑瘤作用。结果:回输hTCR Vβ8.4修饰的PBMC组(即实验组)SCID小鼠的瘤重:(1.402±0.812) g,瘤体大小:(2.218±1.518) cm3; 未经hTCR Vβ8.4基因修饰的PBMC组鼠瘤重:(2.940± 1.441) g,瘤体大小:(3.591±1.546) cm3;注射等体积的生理盐水组鼠瘤重:(4.010±1.777) g,瘤体大小:(4.927±4.535) cm3;基因修饰组与PBMC组比较,瘤重抑制率:52.31%。结论:hTCR Vβ8.4基因能增强PBMC的抑瘤作用,为该基因用于肿瘤的临床基因免疫治疗提供可靠的实验数据。

    Study on the tumor inhibitory effect of T cell transfected hTCR Vβ 8.4 gene in SCID mice

    Abstract Objective: To study the tumor inhibitory effect of human peripheral blood mononuclear cell (PBMC) transfected hTCR Vβ8.4 gene in SCID mice. Methods: The animal model was established by implanting hepatocellular carcinoma cells BEL-7402 subcutaneously in SCID mice. The hTCR Vβ8.4 recombinant plasmid was transfected to health donors PBMC,and then the decorated PBMC was injected in SCID mice through vein.Two weeks later, the mice were killed, observed the average tumor weight and size of each group and tumor inhibitory rate in different groups. Results: The average tumor weight of injecting Saline group was (4.010±1777) g and the average tumor size was (4.927±4.535)cm3 ; The average tumor weight of only injecting PBMC group was (2.940±1441) g, the average tumor size was (3.591±1.546) cm3;The average tumor weight of experiment group was (1.402±0.812) g; Compared with PBMC or Saline group, P<0.05; the average tumor size was (2.218±1.518) cm3;Compared with PBMC or Saline group, P<0.05; The tumor inhibitory rate was 52.31%. Conclusion:The tumor inhibitory effect was reinforced by hTCR Vb8.4 gene, providing reliable experiment data about hTCR Vb8.4 gene used in preclinical of gene immunization treatment. ......
