马齿苋总黄酮抗小鼠缺氧作用及其机制研究

     [摘要] 目的:筛选马齿苋抗缺氧作用有效成分,并对筛选出的有效成分进行作用机制的研究。方法:马齿苋抗缺氧作用有效成分筛选:将70只小鼠随机分为总黄酮组、总生物碱组、多糖组、酸性成分组、碱性成分组、人参总皂苷组(阳性对照)和生理盐水组(阴性对照),每组10只。各组小鼠予以相应药物灌胃1周后,进行常压缺氧暴露生存时间实验,筛选出马齿苋抗缺氧作用的有效成分。马齿苋总黄酮抗缺氧机制研究:将90只小鼠随机分为总黄酮组、人参总皂苷组和生理盐水组,各组再分成3个亚组,即缺氧暴露12、24和36 h组,每组10只。将小鼠置于氮氧混合气(10% O2+90% N2)舱内分别缺氧暴露12、24和36h后,测定血红细胞计数、血红蛋白含量、血细胞压积、血浆红细胞生成素(erythropoietin, EPO)以及肾脏与大脑皮质EPOmRNA的表达水平。结果:与生理盐水组比较,马齿苋总黄酮组小鼠缺氧暴露生存时间明显延长;马齿苋总黄酮组小鼠不同缺氧时段肾脏和大脑皮质EPO mRNA的表达水平明显增加,血浆EPO、红细胞计数和血红蛋白含量亦明显升高。结论:总黄酮是马齿苋抗缺氧作用的有效成分,其延长小鼠缺氧生存时间的机制可能是促进EPO的表达以及红细胞和血红蛋白的生成。

    Effects of flavones extracted from Portulaca oleracea on ability of hypoxia tolerance in mice and its mechanism

    ABSTRACT Objective: To identify antihypoxia ingredients extracted from Portulaca oleracea and to find out the possible mechanism of its antihypoxia actions. Methods: Seventy mice were randomly divided into seven groups which were untreated (normal saline), ginsenosidestreated, polysaccharidetreated, acidic componentstreated, basic componentstreated, alkaloidstreated and flavonestreated groups, and the ingredients of polysaccharide, acidic components, basic components, alkaloids and flavones were extracted from Portulaca olerace. The mice in each group were fed with corresponding ingredients for one week respectively. Then the survival time of mice in hypoxic conditions was observed. Another 90 mice were divided into 3 groups: untreated (normal saline), ginsenosidestreated and flavonestreated groups. The mice in each of these 3 groups were divided into 3 subgroups according to 12, 24 and 36hour exposure to hypoxia (10% oxygen and 90% nitrogen), respectively. After exposure to hypoxia, the red blood cell count (RBC), hemoglobin (Hb) concentration and hematocrit (HCT) in mice were determined. The plasma erythropoietin (EPO) levels of mice were detected by enzymelinked immunosorbent assay (ELISA) and the relative values of EPO mRNA in renal tissue and pallium of mice were determined by reverse transcriptasepolymerase chain reaction (RTPCR). Results: The survival time of mice in hypoxic conditions in flavonestreated group was significantly longer than that in the untreated group. The RBC, Hb concentration, HCT, plasma EPO level and the relative values of EPO mRNA in renal tissue and pallium of mice were significantly higher in the flavonestreated group than those in the untreated group. Conclusion: The antihypoxia ingredients extracted from Portulaca oleracea are flavones and the antihypoxia effects may be obtained by improving the expression level of EPO and accelerating the generations of erythrocyte and Hb. ......
