Humanin的基因克隆及序列测定

    摘要:目的 利用基因工程方法对Humanin进行基因克隆并测定其序列,为进一步对阿尔茨海默病进行基因治疗奠定基础。方法 采用非对称互补引物/模板法,制备两端含有酶切位点的Humanin的cDNA,将其克隆入pVAXI载体中并测序。结果 经酶切鉴定、测序分析,表明所插入的基因片断为Humanin基因,与设计完全相同。结论 成功地克隆了Humanin基因。

    The cloning and sequencing of HumaninJin Hui, Wang Weixi, Yang Guangxiao, Wang Quanying, Hu Haitao, Han Taizhen

    ABSTRACT: Objective To use gene engineering technique to clone and sequence theHumanin. Methods By means of asymmetrical primer/template, double stranded cDNA of Humanin was constructed, which had restriction enzymes sites on the two extremes. Then the cDNA was subcloned into the plasmid pVAXI and sequenced. Results Evidences of DNA sequence analysis and restriction enzymes digestion showed that the inserted fragment wasHumanin cDNA, consistent with the fragment we designed. Conclusion The Humanin cDNA was cloned successfully. ......
