新型骨替代材料-珍珠层的生物相容性研究

摘 要:[目的]研究珍珠层的生物相容性。[方法]在体外培养的第3代成骨细胞中分别加入珍珠层(实验组)、橡胶(对照组)的材料浸提液,观察细胞形态、细胞膜和超微结构的变化;通过BrdU核掺入法检测细胞增殖情况、通过MTT法检测成骨细胞代谢活性。[结果]实验组细胞形态及细胞结构良好、细胞膜完整;对照组中细胞形态发生改变,细胞膜及胞浆内细胞器均有破坏;BrdU及MTT检测结果显示珍珠层对人成骨细胞的增殖和代谢活性无阻滞作用。[结论]珍珠层具有良好的生物相容性,可作为骨替代材料应用于骨缺损修复。关键词:骨替代材料; 珍珠层; 成骨细胞; 生物相容性

Abstract:[Objective]To study the biocompatibility of nacre on cultured human osteoblasts in vitro[Method]The iliac bone of human were added with collagenasetrypsin,a lot of osteoblasts were released outThen the osteoblasts were purified and culturedSubsequently,the 3rd generation cultured human osteoblasts were inoculated with the extract of three materials:nacre,hydroxyapatite,and rubberThe appearance of osteoblast was observed under the phase contrast microscopeMicrostructure of the osteoblast was observed under transmission electron microscopeAdditionally,to determine whether these three materials affect cell proliferation,the numbers of BrdUlabeled cells were examined at three groupsMTT assay was tested to observe the viability of human osteoblasts[Result]Under the phase contrast microscope,the appearance of the osteoblasts in two groups were spindle or triangle like,and black nodes were seen in confluent cell layer 19 days after culturedThe observation of electron microscopy showed that the nucleus of the osteoblasts in two groups were big and round,and there were a large number of mitochondrion and rough endoplasmic reticulum in two groupsStereological analyses of the numbers of BrdUlabeled cells revealed that females produced more cells than males in the dentate gyrus but not in the subventricular zone[Conclusion]The nacre have no harmful effect to character of biology of human osteoblasts in vitro ......
