羟基喜树碱对人骨髓增生异常综合征MUTZ1细胞体外作用的研究

     [摘要]

    目的观察羟基喜树碱对人骨髓增生异常综合征(MDS)MUTZ1细胞体外生长的影响。方法采用四甲基偶氮唑蓝(MTT)比色法，测定羟基喜树碱对人骨髓增生异常综合征MUTZ1细胞生长的抑制作用，并绘制浓度抑制率曲线；采用流式细胞技术检测药物诱导的凋亡细胞量；光镜及透射电镜观察羟基喜树碱处理后细胞形态学的变化。结果羟基喜树碱对MUTZ1细胞生长抑制作用呈现剂量和时间依赖性，其作用不同时间的半数抑制浓度(IC50)分别为91198、8.983、6204、2509、0364。羟基喜树碱可诱导MUTZ1细胞凋亡，呈现剂量依赖性。光镜下可见凋亡细胞胞膜完整、胞体固缩、核固缩、核染色质碎裂；透射电镜下可见凋亡细胞核染色质凝聚、边集，部分细胞核膜消失，细胞浆浓缩、密度增加，浆内可见大小不规则的染色质团块。结论羟基喜树碱对MUTZ1细胞生长有明显的抑制作用，并可诱导MUTZ1细胞凋亡。

    Abstract:Objective To study the effect of hydroxycamptothecine(HCPT) on MUTZ1 cell line of myelodysplastic syndromes(MDS). Methods We treated MUTZ1 cells with HCPT, detected the proliferation inhibition by MTT. The cells apoptosis induced by HCPT was analyzed with flow cytometry(FCM). Apoptotic morphological features was observed by microscope and transmission electronmicroscope. Results HCPT could inhibit the growth of MUTZ1 cells in doseand timedependent manners. Fifty percent inhibitory concentration(IC50) of HCPT for different time point was 91.198,8.983,6.204,2.509,0364 respectively.
