赤芍中芍药苷提取溶剂的选择

     摘要:目的探讨赤芍药苷提取的最佳溶剂。方法分别用甲醇、75%乙醇、重蒸馏水提取赤药中芍药苷,用 YWG-C18(250×4.6 mm)色谱柱,以甲醇-0.05 mol/L磷酸二氢钾溶液(32:68)为流动相测定芍药苷,检测波长为230 nm。结果在所试浓度(0.010~0.200 μg/μl)范围内线性关系良好 r=0.999 8,n=5;以甲醇、75%乙醇、重蒸馏水作溶剂赤芍苷的含量分别为3.01%,3.09%,3.07%。结论结果表明,以甲醇、75%乙醇、重蒸馏水作溶剂提取赤芍中芍药苷的含量无显著性差异(P>0.05);使用蒸馏水作溶剂可避免甲醇较强的毒性且廉价;我们认为使用重蒸馏水做溶剂优于乙醇、甲醇。

    Effects of Different Solvents on the Extraction of Paeoniflorin from Radix Paeoniae Rubra

    AbstractObjectiveTo select the optimal solvent to extract paeoniflorin from Radix Paeoniae Rubra.MethodsPaeoniflorin was extracted from Radix paeoniae Rubra with methanol,75% ethanol and redistilled water ,respectively .Paeoniflorin was separated on a YWG-C18(250 mm×4.6 mm) using methanol:0.05 mol/L KH2PO4(32:68) as a mobile phase and determined at 230 nm.ResultsThe linear curve was ideal within the tested range of 0.010~0.200 μg/μl(r=0.9998,n=5);the average content of paeoniflorin extracted with methanol,75% ethanol and redistilled water was 3.01%,3.09%,3.07%,respectively.ConclusionThere was no significant difference in extraction of paeoniflorin with these three solvents(P>0.05),whereas methanol is highly toxic,redistilled water is a low-priced and safe solvent,indicating ettraction with redistilled water is better. ......
