-80℃冻存外周血造血干细胞的应用研究

     摘要:目的:探求一种操作简便实用、造血干细胞回收率和存活率高、成本低的外周血造血干细胞的冻存方法。方法:CP-1和5%白蛋白作保护剂在–80℃条件下直接冻存,并检测冻存前后外周血干细胞的活性,单个核细胞(MNC)和CFU-GM、CD34+细胞的回收率。结果:冻存前后外周血干细胞活性,单个核细胞(MNC)和CFU-GM、CD34+细胞的回收率无明显差异(P>0.05),输注后造血均重建。结论:CP-1和5%白蛋白作保护剂在–80℃条件下直接冻存干细胞是一种简便、可靠、安全、成本低的外周血干细胞冻存方法,具有广泛的临床推广价值。

    Study on the Freezing Procedure to Store Peripheral Blood Stem Cells at -80℃

    Abstract:Objective:To seek a convenient, practical freezing procedure with high recovery rate and survival rate as well as low cost to store peripheral blood (PB) stem cells. Method:The PB stem cells were froze at–80℃ directly with CP-1 and 5% albumin as protective agents. The activity of PB stem cells and recycle rate of mononuclear cell (MNC), CFU-GM and CD34+ cells before and after freezing were detected. Result:The activity of PB stem cells and recovery rate of MNC, CFU-GM and CD34+ cells have no significant differences before and after freezing(P>0.05). Hemopoiesis could be reconstructed after infusion all. Conclusion:Freezing stem cells directly at –80℃ by using CP-1 and 5% albumin as protective agents is a convenient, reliable, safe and low cost procedure to store PB stem cells, which has the possibility to be used in clinical widely. ......
